Our AI-supported INCYPROsitefinder algorithm analyzes the protein’s 3D structure to identify optimal cysteine positions for crosslinking. These sites are chosen to support the active conformation while avoiding critical binding regions.
A panel of tris-armed crosslinkers offer different reactivities, linker lengths, and hydrophilicity. These small molecules lead to only minimal modification of the protein.
The designed triple-cysteine protein variant is reacted with the crosslinker, selectively producing a defined INCYPRO protein and effectively stabilizing the protein.
